مطالعه تنوع ژنتیکی، توانایی گرهزایی و کارایی تثبیت نیتروژن باکتریهای ریزوبیوم جداسازی شده از گرههای یونجه
تثبیت نیتروژن مولکولی توسط باکتریهای ریزوبیومی، منبع اصلی برای ورود پایدار نیتروژن مورد نیاز گیاهان به سیستمهای کشاورزی است. تثبیت زیستی مولکول نیتروژن در کشاورزی میتواند به وسیله مایهزنی گیاهان لگوم با باکتریهای ریزوبیومی دارای توانایی بالا در تثبیت این مولکول بهبود پیدا کند. آگاهی از تنوع زیستی جمعیتهای بومی این باکتریها برای طراحی تدابیر مایهزنی موفق میتواند مفید واقع شود. در این تحقیق تنوع زیستی، توانایی گرهزایی و کارایی تثبیت نیتروژن 48 باکتری ریزوبیوم جداسازی شده از گرههای گیاه یونجه کشت شده در مناطق مختلف استان همدان مورد مطالعه قرار گرفت. نتایج بررسی تنوع ژنتیکی سویههای مورد مطالعه با استفاده از تکنیک ITS-PCR-RFLP نشان داد که باکتریهای مورد بررسی دارای تنوع ژنتیکی قابل توجه بوده و در سطح شباهت 70 درصد به 4 گروه III ,II ,Iو IV تقسیمبندی شدند.نتایج آزمون گرهزایی نشان داد که اکثر سویهها دارای توانایی ایجاد گره در گیاه یونجه بودند، در حالیکه در اثر تلقیح این گیاه با سویههای KH16، KH24، KH10، KH6، KH133 و KH193 گرهای حاصل نشد. بر اساس نتایج حاصل از آزمون بررسی کارایی تثبیت نیتروژن سویههای مورد مطالعه، این باکتریها به چهار دسته غیر مؤثر، نسبتاً مؤثر، مؤثر و خیلی مؤثر تقسیمبندی شدند.

اثر قارچهای میکوریز آربسکولار بر رشد و جذب عناصر غذایی پایه نارنج در شرایط تنش کم آبی
در مناطق خشک و نیمه خشک کمبود آب سبب کاهش تولیدات کشاورزی میشود. مرکبات کشور در سالهای اخیر در پی خشکسالیهای مستمر با کاهش تولید و افت سطح باغات مواجه است. قارچهای میکوریز آربسکولار با مکانیسمهای مانند افزایش جذب عناصر غذایی به کاهش اثرات کم آبی در گیاهان میزبان کمک میکند. در این پژوهش اثر قارچ گلوموس موسه و گلوموس ورسیفرم بر رشد و جذب عناصر غذایی در پایه نارنج در یک آزمایش گلخانهای در خاک استریل بررسی گردید. آزمایش به صورت فاکتوریل در قالب طرح پایه کاملاً تصادفی با سه فاکتور شامل قارچ میکوریز آربسکولار در سه سطح شامل گلوموس موسه و گلوموس ورسیفرم وشاهد، کم آبی در 4 سطح (دورهای آبیاری 2، 4، 6 و 8 روز)و پایه نارنج در سه تکرار صورت گرفت. کم آبی میانگین وزن خشک ریشه و اندام هوایی و جذب عناصر غذایی اندام هوایی و ریشه (فسفر، نیتروژن، آهن، مس و منگنز) را کاهش، در حالی جذب روی اندام هوایی و ریشه را افزایش داد.میزان وزن خشک ریشه و اندام هوایی، جذب فسفر، نیتروژن، آهن، منگنز، مس و روی در اندام هوایی پایه نارنج تلقیح شده با قارچ میکوریزی نسبت به پایه غیرمیکوریزی در شرایط تنش کم آبی، بالاتر بود. درصد کلنیزاسیون ریشه در تیمارهای دارای قارچ بیشتر از تیمارهای بدون قارچ بود. در پایه نارنج تلقیح شده با قارچ میکوریزی با افزایش تنش کم آبی درصد کلنیزاسیون ریشه کاهش یافت.

ماکروفونهای ساکن در خاک از اجزای مهم تنوعزیستی خاک بهشمار میروند. با هدف بررسی تأثیر نوع محصول و شیوه مدیریت زراعی بر جامعه ماکروفون خاک سه زیستگاه شامل مزارع یونجه، مزارع کمنهاده گندم و مزارع پرنهاده گندم مورد پیمایش قرار گرفت. برای هر زیستگاه شش واحد نمونهگیری منظور گردید. در هر واحد نمونه گیری ماکروفونهای خاک با استفاده از تلههای چالهای جمع آوری و به تفکیک خانواده شمارش شدند. تجزیه و تحلیل داده ها با استفاده از آنالیزهای تقابل، تشابه و تجزیه به مولفههای اصلی انجام گرفت. نتایج آنالیز تقابل نشان داد که شاخص شانون در محصول یونجه (11/2) بیش از محصول گندم ( 88/1) میباشد. انجام آنالیز تشابه حاکی از اختلاف در ترکیب جامعه ماکروفون خاک دو محصول داشت به نحوی که ماکروفون های مفید شامل عنکبوتیان، سوسک زمینی و کرمهای خاکی در محصول یونجه فراوان تر بودند. پایائی و تخریب کمتر محصول یونجه عامل اصلی این فراوانی تشخیص داده شد. در حالی که اختلاف محسوسی بین دو شیوه مدیریت کمنهاده و پرنهاده گندم از نظر شاخصهای تنوع مشاهده نگردید با این وجود ترکیب متفاوتی از ماکروفونها در دو شیوه مدیریتی شکل گرفت. گرایش ماکروفونهای مفید خاک به سکونت در زیستگاه کمنهاده قابل درک بود. عمده اختلاف در ترکیب ماکروفونها به عدم کاربرد علفکشها و مصرف کمتر کودهای نیتروژنه در زیستگاه کمنهاده گندم نسبت داده شد. به طور کلی نتیجه گرفته شد که بهبود تنوعزیستی ماکروفون های خاک نیازمند بکارگیری مدیریت کمنهاده و وارد کردن بقولات در برنامه تناوب زراعی میباشد.

تأثیر قارچکش کربوکسین تیرام (Carboxin Thiram) بر رابطه همزیستی قارچهای میکوریزا- آربسکولار با گیاه گندم
کودهای بیولوژیک علاوه بر اثرات مثبت که بر حاصلخیزی پایدار خاک دارد، از جنبههای اقتصادی و زیست محیطی نیز مثمرثمر واقع شده و میتوانند به عنوان جایگزین مناسب و مطلوب برای تمامی یا حداقل بخشی از کودهای شیمیایی باشند. از مهمترین انواع کودهای بیولوژیک میتوان به قارچهای آربوسکولار میکوریزی اشاره کرد. با توجه به گستردگی استفاده از قارچکش Carboxin Thiramبرای کنترل عوامل بیماریزای قارچی در کشت گندم و به منظور تعیین میزان تأثیر این قارچکش در برقراری رابطه همزیستی گیاه گندم با گونههای مختلف قارچهای میکوریز آربسکولار، این پروژه طراحی و اجراء گردید. با انجام آزمون گلخانهای، تأثیر قارچکش Carboxin Thiram در دو سطح مصرف (به میزان دو در هزار) و عدم مصرف بر همزیستی شش گونه از قارچهای میکوریزی (Glomus clarum Glomus caledonium ، Glomus mosseae ،sp Glomus ، Glomus geosporum، Glomus versiforme) بر شاخصهای رشد گیاه گندم رقم چمران و همچنین بر روی تعداد اسپور و درصد کلونیزاسیون ریشه گیاه با این قارچها بررسی گردید. نتایج تحقیق نشان داد که استفاده از این قارچکش به صورت بذرمال تأثیر معنیداری در هیچیک از اجزاء عملکردی گیاه در بر نداشت. تلقیح با قارچ میکوریز و استفاده از Carboxin Thiram تعداد اسپور (p<0.01) و درصد کلونیزاسیون ریشه (p<0.05) را به طور معنیداری افزایش داد. بیشترین تعداد اسپور تولیدی در همزیستی قارچ Glomus geosporum و در سطح استفاده از Carboxin Thiram با 37/34 اسپور به ازاء هر گرم خاک و کمترین میزان در قارچ G.clarum با 5 اسپور در هر گرم خاک مشاهده شد. نتیجه کلی حاصل از این پژوهش حاکی از عدم تأثیر منفی بکارگیری بذور گندم حاوی قارچکش Carboxin Thiramدر برقراری رابطه همزیستی میکوریزی با گونههای مورد استفاده در این تحقیق میباشد.

با توجه به روند افزایشی توسعه اراضی شور و کمبود اراضی زراعی مطلوب برای کشاورزی شناسایی راهکارهایی که باعث افزایش مقاومت این گیاهان در برابر شرایط شور میشود از اهمیت زیادی برخوردار است. استفاده از باکتریهای محرک رشد یکی از این راهکارها محسوب میشود. بنابراین به منظور بررسی تأثیر چند گونه از باکتریهای محرک رشد و ترکیب این گونهها در شرایط تنش شوری بر شاخصهای رشد دو رقم گندم، آزمایشی به صورت فاکتوریل در قالب طرح کاملاً تصادفی با 3 تکرار انجام شد. فاکتورهای آزمایش شامل تنش شوری در سه سطح) 335/0 (شوری آب آبیاری به عنوان شاهد)، 6 و 14 دسیزیمنس بر متر (که از ﻃﺮﻳﻖ اﺿﺎﻓﻪ ﻧﻤﻮدن نمک به آب ﺑﺎ 335 EC ﻣﻴﻜﺮوزﻳﻤﻨﺲ ﺑﺮ ﻣﺘﺮﺗﻬﻴﻪ ﮔﺮدﻳﺪ)( و فاکتور باکتریهای PGPR در هشت سطح شامل 1- (تیمار بدون باکتری به عنوان شاهد (Control)) 2- (آزوسپیریلوم لیپوفروم سویه of) 3- (ازتوباکتر کروکوکوم سویه 5) 4- (سودوموناس فلورسنس سویه 169) 5- (آزوسپیریلوم لیپوفروم سویه of+ ازتوباکتر کروکوکوم سویه 5) 6- (آزوسپیریلوم لیپوفروم سویه of + سودوموناس فلورسنس سویه 169) 7- ( ازتوباکتر کروکوکوم سویه5 + سودوموناس فلورسنس سویه 169) و 8 - (آزوسپیریلوم لیپوفروم سویه of + ازتوباکتر کروکوکوم سویه 5 + سودوموناس فلورسنس سویه 169) درگلخانه موسسه تحقیقات خاک و آب و به روش کشت در شن (Sand Culture) انجام شد. ارقام مورد استفاده در آزماش شامل دو رقم کویر (متحمل به شوری) و قدس (حساس به شوری) بودند. نتایج نشان داد که تنش شوری تأثیر معنیداری برشاخصهای مرتبط با ریشه و اندام هوایی دارد (P<0.01)، به نحوی که با افزایش میزان شوری آب آبیاری، میانگین صفات مذکور کاهش یافت. همچنین نتایج بیانگر تأثیر معنیدار و مثبت باکتریهای PGPR به ویژه ترکیب چند باکتری از جنسهای متفاوت در شرایط شور بود P<0.01)). در مجموع میانگین اکثر صفات در نمونههایی که بذرها به وسیله باکتریهای ترکیبی با جنسهای متفاوت تلقیح شده بودند بالاتر بود براساس نتایج حاصل از مقایسه بین ارقام نتایج بیانگر تأثیر بیشتر باکتریها در رقم حساس بوده است. بنابراین.بر اساس نتایج حاصل از این پژوهش مشخص شد که در سطوح شوری مورد مطالعه، تلقیح با باکتریهای منتخب باعث کند شدن روند کاهش رشد در صفات مورد مطالعه میگردد و باکتریهایPGPR توانستند شاخصهای رشد گیاه را در شرایط شور به طور معنیداری بهبود بخشند.

تأثیر دو گونه قارچ آربوسکولار (Glomu. mosseae و G. etunicatum) بر جذب عناصر غذایی و تولید ریزغده در گیاهچههای حاصل از کشت بافت سیبزمینی
در این پژوهش به منظور بررسی اثرات همزیستی قارچ میکوریز در جذب عناصر غذایی، ارتقاء سازگاری گیاهچههای حاصل از کشت بافت سیبزمینی در گلخانه و افزایش راندمان تولید ریزغده در آنها، گیاهچههایی از دو رقم سیبزمینی (رقمهای آگریا و سانته) با دو گونه قارچ میکوریز آربوسکولارا (Glomus.mosseae و G.etunicatum) به صورت مجزا و در مخلوط با هم در قالب آزمایش فاکتوریل با طرح پایه کاملاً تصادفی و در چهارتکرار مایهزنی شدند. مایهزنی در گلخانه و در هنگام انتقال گیاهچهها انجام شد. هشت هفته پس از مایهزنی درصد کلونیزاسیون ریشه در گیاهچهها تعیین شد و از میزان عناصر غذایی آنها ( فسفر، آهن، روی و منگنز) اندازهگیری به عمل آمد. ریزغدههای تولیدی به اندازههای مختلف تفکیک شده و عملکرد کل برآورد شده و درصد ماده خشک ریزغده نیز تعیین گردید. نتایج نشان داد که زاد مایة قارچ بر میزان کلونیزاسیون و تمامی صفات اندازهگیری شده در سطح احتمال 1% اثر معنیدار داشت. اثر نوع رقم صرفاً در مورد آهن و روی تفاوت معنیدار نشان نداد. اما در سایر صفات اندازهگیری شده در سطح احتمال 1% معنیدار شد. با مقایسه میانگین صفات مشخص شد که مایه زنی باگونه G.etunicatumو مخلوط دو گونه بر شدت کلونیزاسیون ریشه، جذب عناصر غذایی و همچنین تعداد ریزغدة تولیدی اثرات مثبتتری نسبت به گونه G. mosseae داشتند. بیشترین میزان جذب فسفر، آهن، روی و منگنز در گیاهچههای مایهزنی شده با مخلوط دو گونه به دست آمد. بیشترین میزان ریزغده در مایهزنی با مخلوط دو گونه قارچ (متوسط تعداد 81/13 عدد ریزغده در گیاهچه) حاصل گردید که نسبت به کاربرد جداگانه دو گونه و تیمار شاهد در سطح 05/0 تفاوت معنیدار داشت. با کاربرد جداگانة دو گونه قارچ و نیز مخلوط آنها درصد ماده خشک ریزغده افزایش پیدا کرد که تفاوتها با تیمار شاهد در سطح 05/0 معنیدار شد. در مجموع مایهزنی گیاهچههای سیبزمینی با دو گونه قارچ میکوریز به صورت جدا و در مخلوط با هم کمک قابل توجهی به جذب عناصر غذایی کرد که به نوبه خود ضمن افزایش زیست توده گیاهی اثرات مثبتی در استقرار گیاهچهها و افزایش راندمان تولید ریزغده در آنها داشت.
